For me, it is the CR-J/CR-SS/CR-N loop between Stockholm, WI and Pepin, WI.

J Cooke, but it's in my back yard so that's and easy one.

In MN my favorite is the twisty section of Hwy 76 between Houston and Caledonia.
In Iowa it is B25.
In WI it is probably SS.
This is subject to change from season to season.

I'm with Mikey on this, jay Cooke is awesome (minus that stupid hair pin that catches me off guard every time). Awesome road, not many Leo's and gorgeous scenery.

I thought someone told me that road was ruined, meaning it was all straightened or something. I haven't been on it in years, but I was in Houston yesterday and wondering whether it was worth a shot. I decided against it. Maybe I should have checked it out. I loved riding it uphill, in a southeasterly direction.

Haven't been down that way this year, but I'll check it out the next time I head to WI.
As for faves, MN-76 is (was) definitely one. Also really like 21 out of Lanesboro. Something about being able to see the next 4 corners that appeals to me.
For WI, there are the standards like Mindoro and Wildcat. I also like the entire length of Cty P from Coon Valley to Rockton. But really, there are too many to name. In a day of riding I'll hit about 50 different roads for a few miles at a time with each section just as good as another, making it hard to pick a favorite. So I'll just blanket the entire area south & west of I-90 and north of the Wisconsin River, as my "favorite".
BTW, I have a detailed map of SW Wisconsin serving as artwork in my garage. ;D

I rode 76 from McGregor to Houston two weeks ago and did not notice any straightening. I think it would be prohibitively expensive to straighten out a road that is only a state highway (that is not as busy as highway 61 along the north shore.)

Everything in Wis. East of CR-D between HWY-10 and the Mississippi all the way to who knows?GJ ::)

I like SS as it has gorgeous scenery and the way the road snakes along the ridge is fun. DD just north of 54 might be a favorite too. The multitude of banked corners on DD are hard to beat. Minduro Cut is great, very technical but not as relaxing as flying through the other roads, so it gets some deductions, however depending on my mood the Cut is also the best road for the same reasons - tight and having fun in 2nd gear tossing the bike around.
My favorite small section of road in MN is County highway 25 off of 61 about 15 miles south of Wabasha. It's only a mile or two long but it is gorgeous and has some of the best sets of turns in the state with great road conditions and views.

I've always liked Hwy 38 from Grand Rapids to Effie. Also a fan of Hwy 65 north of Hwy 1 - for different reasons that twisties, which there are some. Old bridges wood plank bridges, the very short squirt through the Bois Forte reservation is very scenic, and it is extremely remote.
And the former Hwy 1 from Ely to Finland, pre-construction.

This road was outstanding. Too bad they straightend out parts of it. It is still good though.
